The task of biotechnology, environment and toxicology department

The department of biotechnology and toxicology is developing and putting into practice environmental technologies directed at disposal of refinery waste and environmental air protection. It is providing the determination of biological degradability of products and evaluation of their toxic features.

Competence and main activities of biotechnology, environment and toxicology

  • development of technology for the decontamination of soil contaminated by crude oil substances and implementation of the technology
  • investigation and development of polluted air purification by means of bio-filters
  • introducing of new testing methods directed at biologic degradability of substances
  • development of new eco-technologies on the basis of physical and chemical processes.
  •

      A.  Laboratory tests

Determination of non-polar extractible substances by IR spectroscopy

The method is based on crude oil substances extraction with non-polar solvent (Ledon 113). The co-extracted polar or semipolar sobstances are separated through adsorption on silicagel and the extract is analyzed in the IR spectral range.

  • in waste water according to DIN 3814, 4 vol., practically consistent to STN 83 0540 part 4
  • The method is suitable to determine concentrations of hydrocarbons - crude oil substances above 0,1 mg/l.

  • in water extract - according to DIN 3814, 4 vol., practically consistent to STN 83 0540, part 4
  • From the test materials the non-polar extractible substances that are soluble in water are determined at exactly stated conditions. The method is applicable for solid materials, with grain size maximum 1 cm3, pastes and sludges.

  • in soils according to STN 83 0540 part 4, modified by the Centre for evaluation of laboratory capability (ASLAB Prague)
  • in sludges according to STN 75 7952
  • in compost according to STN 75 7952, modified method
  • The method determines the main part of non-polar hydrocarbons in sludges, with a weight fraction above 1g/kg in dry matter of sludge.

      B.  Special test methods

Gene mutation (Ames test - OECD 471)

The test is done to determine gene mutation caused by the studied substances on bacterial strains of Salmonella typhimurium, with and without metabolic activation.

Chromosome mutation in vitro (Micro-nucleus test - OECD 474)

The test is for the detection of chromosome mutation caused through the effect of studied substance, on lymphocytes of the peripheral human blood, with and without metabolic activation.

DNA damage and repair (Reparation test - OECD 482)

The test serves for the detection of DNA damage and repair caused by the investigated substance on bacterial strains of Escherichia coli with and without metabolic activation.

    Biodegradability of products (according to method CEC L - 33 - A - 93)

The method states biologic degradability of various water insoluble crude oil products.
